脳循環
私たちの実験班では、ウサギを用いて、いろいろな薬剤の脳循環に与える影響を、クラニアルウインドウ法を使用して検討しています。
この方法は、ウサギの頭頂骨に穴を開け、硬膜を剥離し、脳軟膜の血管を露出させ、直視下に血管を観察することにより、各薬剤の脳血管に与える影響を検討する方法です。
頭頂の開頭部に、カバーガラスのついたリングを歯科用セメントで固定し、リングに設置したポートより人工髄液を還流します（クラニアルウインドウの作成）。
さらに薬液注入用のポートより、いろいろな薬剤を投与することによる血管径の変化を、ビデオマイクロメータで計測します。
この方法の特徴は、脳に直接薬剤を投与するので、全身投与した際に起こる様々な影響を排除した直接的効果を検討できることです。
最近では、ウサギの大動脈弓を露出し、腕頭動脈、総頚動脈、鎖骨下動脈をクランプすることによる全脳虚血モデルを作成することで、虚血前後の薬剤の与える影響を検討しています。
